It was founded by the Catholic Monarchs in 1504 to treat the wounded from the War of Granada. Building work began following a design by Enrique Egas (late Gothic), although Charles V finished the project in Renaissance style.

The façade was not finished until the first half of the 17th century.

It is currently the main headquarters of the University of Granada, home to the Rectory and other university services.
